IL-12 is a 70 kDa heterodimeric cytokine composed of two covalently linked chains, p40 and p35, and is secreted by activated B cells and macrophages. IL-23 is a structurally and biologically-related cytokine comprised of the same p40 subunit and a p19 subunit. For optimal detection of total IL-12 p40 (monomer, heterodimer, homodimer) by ELISA, a combination of purified C15.6 antibody for capture and biotinylated C17.8 antibody for detection is recommended. Fluorochrome conjugated C15.6 antibody is effective for staining intracellular IL-12.
